+= does not do appending, it does addition. "diffopt" is not a vim variable, it is option and & is a sigil denoting option. There is no appending taking place here: you need

    let &diffopt.=(empty(&diffopt)?'':',').$VIMDIFFOPT

(.= is appending). You should never confuse :let and :set.
